Incidence de la salinite sur les equilibres de distribution hydrique et ioniques et sur le metabolisme respiratoire dans le muscle blanc, le muscle rouge et la branchie chez le muge Chelon labrosus ArchiMer
Gallis, J; Piquemal, S; Lambert, J; Sergent, P; Rousseau, V.
Oxygen consumption, water, K super(+) and Na super(+) contents were measured in white muscle, red muscle and gill epithelium in the mullet Chelon labrosus) in relation with the duration of fresh water or sea water (1500mOsm) adaptation. The unit respiratory activities in gill and red muscle where respectively 5 and 3 timesmore elevated than in white muscle. The pericellular K super(+)/Na super(+) ratio of the 3 tissues decreased with the fresh water adaptation duration. In the gill, the intracellular K super(+)/Na super(+) ratio was stable while the parameter decreased strongly in the white muscle. The results are discussed in relation to energetic cost of homocellular regulation and homoepithelial regulation.

Differential distribution of V-type H+-ATPase and Na+/K+-ATPase in the branchial chamber of the palaemonid shrimp Macrobrachium amazonicum ArchiMer
Boudour-boucheker, Nesrine; Boulo, Viviane; Charmantier-daures, Mireille; Grousset, Evelyse; Anger, Klaus; Charmantier, Guy; Lorin-nebel, Catherine.
V-H+-ATPase and Na+/K+-ATPase were localized in the gills and branchiostegites of M. amazonicum and the effects of salinity on the branchial chamber ultrastructure and on the localization of transporters were investigated. Gills present septal and pillar cells. In freshwater (FW), the apical surface of pillar cells is amplified by extensive evaginations associated with mitochondria. V-H+-ATPase immunofluorescence was localized in the membranes of the apical evaginations and in clustered subapical areas of pillar cells, suggesting labeling of intracellular vesicle membranes. Na+/K+-ATPase labeling was restricted to the septal cells. No difference in immunostaining was recorded for both proteins according to salinity (FW vs. 25 PSU). In the branchiostegite,...

Salinity tolerance, ontogeny of osmoregulation and zootechnical improvement in the larval rearing of the Caledonian Blue Shrimp, Litopenaeus stylirostris (Decapoda, Penaeidae) ArchiMer
Pham, Dominique; Charmantier, Guy; Wabete, Nelly; Boulo, Viviane; Broutoi, Francis; Mailliez, Jean-rene; Peignon, Jean-marie; Charmantier-daures, Mireille.
The ontogeny of osmoregulationwas investigated in Litopenaeus stylirostris by studying salinity tolerance and osmoregulatory capacity. Shrimp at different larval and postlarval stages were exposed to various salinities and survival was monitored for 24 h. Survival rates exceeded 80% at salinity over 25 ppt (750 mOsm.kg(-1)) at all the stages. At salinities below to 25 ppt, salinity tolerance was higher in nauplii and zoeae than in mysis larvae. Postlarval stages were able to withstand lower salinities, e.g. 6.0 ppt (176 mOsm.kg(-1)) at PL9 stage, but they were more sensitive than larvae to salinities over 35 ppt (1035 mOsm.kg(-1)). Zoea and mysis larvae slightly hyper-regulated at all tested salinities. After metamorphosis, postlarvae progressively...

Physiological and biochemical changes of wild and cultivated juvenile pink shrimp Farfantepenaeus duorarum (crustacea: penaeidae) during molt cycle ArchiMer
Luis Bonilla-gomez, Jose; Chiappa-carrara, Xavier; Galindo, Carmen; Jeronimo, Gilberto; Cuzon, Gerard; Gaxiola, Gabriela.
Changes in metabolite levels in hemolymph and hepatopancreas were used as indicators of physiological status of juvenile wild and cultivated Farfantepenaeas. duorarum (Burkenroad, 1939), during hyper-osmoregulatory conditions (salinity 22 ppt, 726 mOsm/kg). We analyzed the relationship between biochemical changes of wild and cultivated shrimp at different molt stages by measurement of osmotic capacity, hemocyanin, acylglycerides, cholesterol, glucose, glycogen and total protein. Biochemical analyses of hemolymph (blood) and hepatopancreas showed a decreasing trend in stages closer to molt and an increasing trend before intermolt. Osmotic capacity and blood proteins indicated a significant effect of the molt stage. Stages A. D1' and D''' showed the greatest...

Physiological basis of interactive responses to temperature and salinity in coastal marine invertebrate: Implications for responses to warming ArchiMer
Torres, Gabriela; Charmantier, Guy; Wilcockson, David; Harzsch, Steffen; Giménez, Luis.
Developing physiological mechanistic models to predict species’ responses to climate‐driven environmental variables remains a key endeavor in ecology. Such approaches are challenging, because they require linking physiological processes with fitness and contraction or expansion in species’ distributions. We explore those links for coastal marine species, occurring in regions of freshwater influence (ROFIs) and exposed to changes in temperature and salinity. First, we evaluated the effect of temperature on hemolymph osmolality and on the expression of genes relevant for osmoregulation in larvae of the shore crab Carcinus maenas. We then discuss and develop a hypothetical model linking osmoregulation, fitness, and species expansion/contraction toward or away...

Tolerance of disease‐vector mosquitoes to brackish water and their osmoregulatory ability ArchiMer
Kengne, Pierre; Charmantier, Guy; Blondeau‐bidet, Eva; Costantini, Carlo; Ayala, Diego.
Salinity tolerance is an important trait that governs the ecology of disease‐vector mosquitoes by determining their choice of larval habitat, and consequently their ecological and geographical distribution. Here, we used laboratory strains to determine the osmotic responses of larvae of obligate freshwater disease‐vector mosquitoes (Aedes aegypti, Aedes albopictus, Anopheles coluzzii, An. gambiae, Culex pipiens, and Cx. quinquefasciatus) and assessed their relationship with salinity tolerance. First, we analyzed the acute dose–mortality response of fourth‐instar larvae to salinity; then, we measured their hemolymph osmolality after 24‐h exposure to varying salinities. We found that Ae. albopictus was the most tolerant species, followed by An. coluzzii,...

Ontogeny of osmoregulation in the Pacific blue shrimp, Litopenaeus stylirostris (decapoda, penaeidae): Deciphering the role of the Na+/K+-ATPase ArchiMer
Pham, Dominique; Charmantier, Guy; Boulo, Viviane; Wabete, Nelly; Ansquer, Dominique; Dauga, Clement; Grousset, Evelyse; Labreuche, Yannick; Charmantier-daures, Mireille.
The role of the main ion transporting enzyme Na +/K + − ATPase in osmoregulation processes was investigated in Litopenaeus stylirostris. The development and localization of the osmoregulation sites were studied during ontogenesis by immunodetection of Na+ K+-ATPase using monoclonal antibodies and transmission electron microscopy (TEM). Osmoregulation sites were identified as the pleurae and branchiostegites in the zoeae and mysis stages. In the subsequent post-metamorphic stages the osmoregulatory function was mainly located in the epipodites and branchiostegites and osmotic regulation was later detected in the gills. The presence of ionocytes and microvilli in these tissues confirmed their role in ionic processes. The complete open reading frame of the...

Change of osmoregulatory and hematological parameters in tilapia (Oreochromis niloticus) after exposure to sublethal mercury concentrations ArchiMer
Handayani, Kiki Syaputri; Soegianto, Agoes; Lignot, Jehan-hervé.
The effects of Hg exposure on blood parameters and gill physiology of tilapia (Oreochromis niloticus) were analyzed. Fish maintained in freshwater were exposed for 7 days (d) to sublethal mercury concentrations (0.1 and 1 mg/L). Blood serum osmolality (SO), sodium (Na+), potassium (K+) and chloride (Cl−) ionic concentrations, and hematological parameters were assessed after 1 up to 7 d of exposure. Serum osmolality and ionic concentrations of exposed fish appeared differently affected throughout the experimental period compared to the controls. Osmolality was reduced at the 2 tested concentrations but Na+ and Cl− contents were only altered at 1 mg/L of Hg after 1 d of exposure and values rapidly returned to the control values thereafter. K+ content was...

A transcriptomic approach of salinity response in the euryhaline teleost, Dicentrarchus labrax ArchiMer
Boutet, Isabelle; Ky, Chin Long; Bonhomme, Francois.
Euryhaline teleosts possess the capacity to osmoregulate under various environmental conditions (freshwater to hypersaline water). This physiological capacity is generally monitored using enzyme activity assays (Na+/K+-ATPase...), hormones quantification (prolactine, growth hormone) or their mRNAs expression. To date, few studies addressed the genetic correlates of adaptation to varying salinity at a molecular level in such fish. In the sea bass Dicentrarchus labrax, genetic differentiation was observed at specific allozyme loci between lagoon- and open-sea populations. In the present study, we investigated transcriptomic response of D. labrax to salt- and freshwater acclimation in two organs involved in osmoregulation, gill and intestine. By using...

Description of the unusual digestive tract of Platax orbicularis and the potential impact of Tenacibaculum maritimum infection ArchiMer
Alix, Maud; Gasset, Eric; Bardon-albaret, Agnes; Noel, Jean; Pirot, Nelly; Perez, Valérie; Coves, Denis; Saulnier, Denis; Lignot, Jehan-hervé; Cucchi, Patricia N..
Background Ephippidae fish are characterized by a discoid shape with a very small visceral cavity. Among them Platax orbicularis has a high economic potential due to its flesh quality and flesh to carcass ratio. Nonetheless, the development of its aquaculture is limited by high mortality rates, especially due to Tenacibaculum maritimum infection, occurring one to three weeks after the transfer of fishes from bio-secure land-based aquaculture system to the lagoon cages for growth. Among the lines of defense against this microbial infection, the gastrointestinal tract (GIT) is less studied. The knowledge about the morphofunctional anatomy of this organ in P. orbicularis is still scarce. Therefore, the aims of this study are to characterize the GIT in...

Inter-individual variability in freshwater tolerance is related to transcript level differences in gill and posterior kidney of European sea bass ArchiMer
L'Honoré, Thibault; Farcy, Emilie; Blondeau-bidet, Eva; Lorin-nebel, Catherine.
Acclimation to low salinities is a vital physiological challenge for euryhaline fish as the European sea bass Dicentrarchus labrax. This species undertakes seasonal migrations towards lagoons and estuaries where a wide range of salinity variations occur along the year. We have previously reported intraspecific differences in freshwater tolerance, with an average 30% mortality rate. In this study, we bring new evidence of mechanisms underlying freshwater tolerance in sea bass at gill and kidney levels. In fresh water (FW), intraspecific differences in mRNA expression levels of several ion transporters and prolactin receptors were measured. We showed that the branchial Cl-/HCO3- anion transporter (slc26a6c) was over-expressed in freshwater intolerant fish,...

Ion uptake pathways in European sea bass Dicentrarchus labrax ArchiMer
Blondeau-bidet, Eva; Hiroi, Junya; Lorin-nebel, Catherine.
Ion uptake mechanisms are diverse in fish species, certainly linked to duplication events that have led to the presence of a multitude of paralogous genes. In fish, Na+ uptake involves several ion transporters expressed in different ionocyte subtypes. In the European sea bass Dicentrarchus labrax, several key transporters potentially involved in Na+ uptake have been investigated in seawater (SW) and following a 2 weeks freshwater (FW) acclimation. Using gel electrophoresis, we have shown that the Na+/H+-exchanger 3 (nhe3, slc9a3) is expressed in gills and kidney at both salinities. Quantitative realtime PCR analysis showed a significantly higher nhe3 expression in fresh water (FW) compared to SW. Its apical localization in a subset of gill ionocytes in...

Molecular characterisation of prolactin and analysis of extrapituitary expression in the European sea bass Dicentrarchus labrax under various salinity conditions ArchiMer
Boutet, Isabelle; Lorin Nebel, C; De Lorgeril, Julien; Guinand, B.
dAlthough prolactin has been demonstrated to be the main hormone involved in adaptation to dilute media in several freshwater teleosts, few studies have been conducted in marine teleosts. In the Mediterranean, the sea bass Dicentrarchus labrax inhabits environments ranging from the open sea to coastal lagoons, where salinity varies greatly. We characterised the prolactin (prl) gene and analysed its expression in two organs (gill and intestine) in D. labrax acclimated to either freshwater or seawater. A 2819 bp long sequence encompassing the prl gene and a part (282 bp) of the promoter were identified, and these comprised 5 coding exons separated by 4 introns. Prolactin was similarly expressed in fresh- and seawater adapted fish, although expression in...

Response of the blue shrimp Litopenaeus stylirostris to temperature decrease and hypoxia in relation to molt stage ArchiMer
Mugnier, Chantal; Soyez, Claude.
The effect of different levels of dissolved oxygen (DO) and a decreased temperature combined with severe hypoxia were studied in the blue shrimp Litopenaeus stylirostris in relation to molt stage. In the first experiment, shrimps were submitted to DO concentrations of 5 to 1 mg l(-1). Osmoregulatory capacity (OC) was measured in shrimps at molt stage C after 6, 24, or 48 h of exposure. No mortality was recorded but a significant negative effect of DO concentration on hypo-OC was observed for DO concentrations below 3 mg l(-1). Osmoregulation improved after 48 h exposure, compared to 6 and 24 h exposure. In a second experiment, temperature was decreased from 28 to 22 degreesC in 24 h, before a severe hypoxia of 0.5+/-0.1 mg O-2 l(-1) was applied....

The ClC-3 chloride channel and osmoregulation in the European Sea Bass, Dicentrarchus labrax ArchiMer
Bossus, Maryline; Charmantier, Guy; Blondeau-bidet, Eva; Valletta, Bianca; Boulo, Viviane; Lorin-nebel, Catherine.
Dicentrarchus labrax migrates between sea (SW), brackish and fresh water (FW) where chloride concentrations and requirements for chloride handling change: in FW, fish absorb chloride and restrict renal losses; in SW, they excrete chloride. In this study, the expression and localization of ClC-3 and Na+/K+-ATPase (NKA) were studied in fish adapted to SW, or exposed to FW from 10 min to 30 days. In gills, NKA-alpha 1 subunit expression transiently increased from 10 min and reached a stabilized intermediate expression level after 24 h in FW. ClC-3 co-localized with NKA in the basolateral membrane of mitochondria-rich cells (MRCs) at all conditions. The intensity of MRC ClC-3 immunostaining was significantly higher (by 50 %) 1 h after the transfer to FW,...

Adaptations biochimiques intestinales chez la truite: Influence de l'aliment et de la salinite sur quelques proprietes membranaires ArchiMer
Leray, C.
The authors report the results of physiological and biochemical experiments which confirm the important role player by fish (Salmo gairdneri ) intestinal mucosa in their osmoregulatory process. The ability of the dietary lipids to modulate this function is described. It was demonstrated that the ionic apical permeability of the enterocytes are related to their membrane lipid composition. The importance of the essential fatty acids is emphasized. The time course changes of the apical permeability are of value in improving sea water adaptation, similar conclusions are reported for Nacl supplementation.

Wastewater bioremediation by mangrove ecosystems impacts crab ecophysiology: in-situ caging experiment ArchiMer
Theuerkauff, Dimitri; Rivera-ingraham, Georgina A.; Lambert, Sophia; Mercky, Yann; Lejeune, Mathilde; Lignot, Jehan-hervé; Sucré, Elliott.
Mangroves are tidal wetlands that are often under strong anthropogenic pressures, despite the numerous ecosystem services they provide. Pollution from urban runoffs is one such threats, yet some mangroves are used as a bioremediation tool for wastewater (WW) treatment. This practice can impact mangrove crabs, which are key engineer species of the ecosystem. Using an experimental area with controlled WW releases, this study aimed to determine from an ecological and ecotoxicological perspective, the effects of WW on the red mangrove crab Neosarmatium africanum. Burrow density and salinity levels (used as a proxy of WW dispersion) were recorded, and a 3-week caging experiment was performed. Hemolymph osmolality, gill Na+/K+-ATPase (NKA) activity and gill...

Osmoregulation and salinity-induced oxidative stress: is oxidative adaptation determined by gill function? ArchiMer
Rivera-ingraham, Georgina A.; Barri, Kiam; Boel, Melanie; Farcy, Emilie; Charles, Anne-laure; Geny, Bernard; Lignot, Jehan-herve.
Osmoregulating decapods such as the Mediterranean green crab Carcinus aestuarii possess two groups of spatially segregated gills: anterior gills serve mainly respiratory purposes, while posterior gills contain osmoregulatory structures. The co-existence of similar tissues serving different functions allows the study of differential adaptation, in terms of free radical metabolism, upon salinity change. Crabs were immersed for 2 weeks in seawater (SW, 37 ppt), diluted SW (dSW, 10 ppt) and concentrated SW (cSW, 45 ppt). Exposure to dSW was the most challenging condition, elevating respiration rates of whole animals and free radical formation in hemolymph (assessed fluorometrically using C-H(2)DFFDA). Further analyses considered anterior and posterior gills...

The Na+/K+/2Cl(-) cotransporter in the sea bass Dicentrarchus labrax during ontogeny: involvement in osmoregulation ArchiMer
Lorin, N; Boulo, Viviane; Bodinier, C; Charmantier, G.
This study combines a cellular and molecular analysis of the Na+/K+/2Cl(-) cotransporter (NKCC) to determine the osmoregulatory role of this protein in different tissues during the ontogeny of the sea bass. We have characterized the complete sequence of the NKCC1 isoform isolated from the sea bass gills and have identified, by immunofluorescence, NKCC1, and other isoforms, within the epithelium of the major osmoregulatory organs. Different (absorptive and secretory) functions have been attributed to this protein according to the tissue and salinity. The effects of short- (1-4 days), medium- (7-21 days) and long (6 months)-term freshwater (FW) adaptations were investigated, in comparison with seawater (SW)-maintained sea bass.
